Testoman's formulation philosophy is to address testosterone production through multiple pathways simultaneously, rather than relying on a single ingredient. Endogenous testosterone is produced in the Leydig cells of the testes in response to stimulation from LH (luteinizing hormone), which is in turn secreted by the pituitary gland in response to GnRH (gonadotropin-releasing hormone) from the hypothalamus. The HPG axis (hypothalamic-pituitary-gonadal) is regulated by multiple factors: chronic stress (elevated cortisol inhibits the HPG axis), micronutrient deficiency (zinc, magnesium, vitamin D3 are cofactors of steroidogenesis), estrogen levels (which negatively feedback to inhibit the HPG axis), and the availability of precursors (cholesterol, LDL). Testoman addresses each of these points with specific ingredients.
D-aspartic acid (DAA): the amino acid that stimulates LH and testosterone release:
D-aspartic acid (DAA) is the D-stereoisomer of asparagine (unlike L-aspartate which enters the Krebs cycle), with unique functions in the neuroendocrine system. In hypothalamic cells, DAA stimulates GnRH release; in the pituitary, it stimulates LH release; in Leydig cells, DAA accumulates and directly stimulates the activity of the StAR protein (Steroidogenic Acute Regulatory protein), which is the limiting protein in the transport of cholesterol into the mitochondria where the steroidogenesis chain begins. The study by Topo et al. (2009), published in Reproductive Biology and Endocrinology, documented that supplementation with 3.12 g/day of DAA for 12 days increased LH levels by 33% and total testosterone by 42% in men with low to moderate baseline testosterone. The effects of DAA are more pronounced in men with functional or subclinical hypogonadism (testosterone <400 ng/dL) and less in men with high baseline testosterone (high-performance athletes).
Ashwagandha (Withania somnifera) 5:1: the adaptogen with the most clinical trials on testosterone:
Ashwagandha root extract (5:1, corresponding to a concentrated extract with a high content of withanolides) has the most well-documented effects on testosterone levels of any adaptogen. The randomized clinical trial by Wankhede et al. (2015), published in the Journal of the International Society of Sports Nutrition with 57 men over 8 weeks, documented that supplementation with 300 mg of KSM-66 (5% withanolides extract) 2x/day resulted in a 15% increase in total serum testosterone levels compared to placebo, associated with greater muscle strength (bench press and leg extension) and lower post-workout muscle oxidative stress. The main mechanism involves reducing cortisol levels (which normally inhibits the HPG axis): Ashwagandha reduces cortisol by 14 to 32% in clinical studies, removing this inhibitor of testosterone production. In addition to its effect on testosterone, Ashwagandha has documented effects on sperm quality (concentration, motility) in men with subclinical infertility.
Maca (Lepidium meyenii) 10:1: libido, vitality, and sexual function without a direct effect on testosterone:
Peruvian Maca root extract (10:1) is one of the most widely used male sexual health supplements globally, with a different evidence profile than DAA and Ashwagandha: Maca improves libido, sexual function, and energy without altering circulating levels of testosterone, estrogen, or SHBG (sex hormone-binding globulin). The mechanism by which Maca improves libido is still poorly characterized but seems to involve specific compounds (benzylglucosinolates, macamides, macaenes) that act on the central nervous system to modulate sexual behavior independently of the endocrine axis. The meta-analysis by Gonzales et al. (2012) documented that Maca significantly improved sexual function and libido in men compared to placebo, without detectable hormonal changes.
L-Carnitine L-Tartrate: support for male fertility and testicular energy metabolism:
