ZMA was originally developed by researcher Victor Conte (BALCO Laboratories) in the 1990s, based on the observation that professional athletes had a high prevalence of subclinical zinc and magnesium deficiency — especially in sports with high sweating and large training volumes. Zinc and magnesium losses through sweat are significant (zinc: 0.5 to 1mg/litre of sweat; magnesium: 5 to 8mg/litre of sweat), and in athletes who train daily with abundant sweating, daily losses of these minerals can easily exceed dietary intake. The combination of the two minerals in a single formula with vitamin B6 (a cofactor for amino acid metabolism and the transport of zinc and magnesium themselves) is the most efficient practical approach for joint replenishment.
Zinc (Zinc Oxide): the mineral for testosterone, immunity and protein synthesis:
Zinc (Zn²⁺) is a cofactor for more than 300 metalloproteins and metalloenzymes — the second most abundant trace mineral in the human body after iron. Its most relevant functions for athletes include: support for testosterone synthesis (zinc is a cofactor for 17β-hydroxysteroid dehydrogenase, the final enzyme in testosterone biosynthesis in Leydig cells, and is necessary for maintaining LH receptors in these cells); muscular protein synthesis (zinc is a cofactor for RNA polymerases and zinc finger transcription factors, essential for genetic expression of protein synthesis); immune function (zinc is essential for T-lymphocyte proliferation and NK cell activity); and wound healing (zinc is a cofactor for matrix metalloproteinases that remodel connective tissue during recovery from muscle and tendon injuries). Zinc deficiency in athletes is associated with reduced serum testosterone, increased susceptibility to respiratory infections, delayed wound healing, and reduced strength performance. The clinical trial by Kilic et al. (2010), published in Neuroendocrinology Letters with intensively training wrestlers, documented that supplementation with 3mg/kg/day of zinc for 4 weeks prevented the reduction in serum testosterone induced by exhaustive exercise compared to the placebo group. Zinc Oxide is a form of zinc with reasonable bioavailability (lower than bisglycinate or gluconate, but effective in adequate doses).
Magnesium (Magnesium Oxide): the mineral for enzymes, ATP, and muscle relaxation:
As extensively described in the Magnesium Bisglycinate section of Marvelous Nutrition, magnesium is a cofactor for more than 300 enzymes and is essential for ATP synthesis, muscle relaxation, and neuromuscular function. In the specific context of ZMA, magnesium has an additional critical role: it is a modulator of NMDA and GABA-A receptors in the central nervous system, with an anxiolytic effect and improvement of sleep quality. The main difference between ZMA and Marvelous's standalone Magnesium Bisglycinate is the form of magnesium: ZMA uses Magnesium Oxide (the most concentrated form of elemental magnesium per gram, but with lower bioavailability — ~4% absorption vs. ~15 to 40% for bisglycinate) and Magnesium Bisglycinate uses the higher-absorption chelate form. For users who are not particularly sensitive to the laxative effect of magnesium oxide and who need a combined ZMA formula at an affordable price, ZMA is the convenient solution; for maximum magnesium bioavailability, Magnesium Bisglycinate is the superior choice.
Vitamin B6 (Pyridoxine Hydrochloride): the synergistic cofactor of ZMA:
Vitamin B6 (pyridoxal phosphate in its active form) is included in the ZMA formula for multiple synergistic reasons. The central argument for nocturnal ZMA use is the hormonal recovery cycle that occurs during deep sleep (SWS — Slow Wave Sleep): the pulsatile secretion of GH (Growth Hormone) during the first episode of SWS is the main nocturnal anabolic stimulus, and both zinc and magnesium are necessary for hormonal signaling during this process.
